I'm almost certain "unarmed defense" is not being checked when an assassin wielding a weapon in his main hand and nothing in his off hand tries to parry an incoming attack with his "off hand".

Post above details the base issue, looks like for your unarmed combat bonuses to come into play (unarmed defense or ironhands) you need both hands free -- or at least your main hand.
